This is the latest in a series of attacks on vaccine teams in the country.
The attack took place in Matta area of Khyber Pakhtunkhwa province, which was once a stronghold of the Pakistani Taliban.
This comes a day after Pakistan launched a week-long nationwide vaccination drive to vaccinate 45 million children.
Local police officer Javed Khan said a team of female polio workers was administering medicine to children in a house when terrorists riding a motorcycle opened fire, killing the officer. He said that a search operation is being conducted to trace and arrest the attackers.
Prime Minister Shehbaz Sharif condemned the attack in a statement and vowed strong action against those responsible.

No group immediately claimed responsibility for the attack, but suspicion is likely to fall on militants who often target health workers and police officers assigned to protect them.
According to health and safety officials, more than 200 polio workers and police officers protecting them have been killed in Pakistan since the 1990s.
Extremists often falsely claim that the vaccination campaign is part of a Western plot to sterilize Muslim children.
Since January, 29 cases of polio have been reported in Pakistan. During the ongoing campaign, more than 400,000 trained workers are going door-to-door to vaccinate children.
Pakistan and neighboring Afghanistan are the only two countries where transmission of wild poliovirus has never been stopped, according to World Health Organization,
